Obtaining and quantification of RPE cells from donor eyes using Celltram Oil Pump (surgical model)

Transplantation of RPE-cells in the retina requires high levels of transplantation technology. It is necessary to apply large number of vital cells to get a long remaining transplantation success. We aimed to optimise the operation technique, considering cell injection under minimising the rate of cell damage, and accurate quantification of obtained RPE cells.
Material and methods: 10 donor eyes (from 5 donors) were taken in less than 48 hours post mortem in the Department of Pathology of Rudolfstiftung Hospital for corneal transplantation and after removing the corneoscleral disc were used for more purposes. Obtaining the RPE cells was carried on using an oil hydraulic microinjection pump (Fa. Eppendorf) Then we carried on retinotomy, mobilising and acquisition of the cells and the following quantification. Results: A total number of 10.000 to 28.000 RPE cells were obtained . The morphologic examination showed significant reduction of the cell damages in this approach in comparison with the conventional method using microinjectional syringes.
Conclusion: Applying oil hydraulic microinjection pump allows a gentle and continuos regulation of injectional pressure and reduction of cell damages. The following application of obtained RPE donor cells (cell cultures) is on going.
