Adhesion of Lens-epithelium-cells on different IOL-Materials: influence of a new surface-modification

Introduction: The adhesion of lens-epitheliun-cells (LEC) on intraocular lenses (IOL) is crucial for the development of posterior capsule opacification (PCO). Aim of this study is to test the adhesion criteria of LEC on different IOL-materials with a surface-coating by Dynasilan®. Methods: 54 IOLs (18 PMMA, 18 Silicone, 18 Acrylic) were tested. 9 IOLs of each material were coated with Dynasilan®. Rabbit LEC in passage 3 were cultivated on each IOL for 72 h (10³ cells). After fixation the cells were counted. LEC on 3 IOLs of each material were tested for the adhesion molecule vinculin by antibody staining.
Results: Dynasilan® -coating reduced the adherent LEC on PMMA-IOL by 72.6% and on Silicone-IOL by 32%. Vinculin-antibody-staining on these these cells showed a reduced fluorescence. The Acrylic-IOL was opaque after the Dynasilan® -coating. The number of LEC was increased by 55%.
Conclusion: In PMMA- and Silicone-IOL the number of adherent LEC was significantly reduced by Dynasilan® -coating. These in vitro results suggest a reduced PCO-rate. In vivo testing has to be performed to varify these results. These preliminary results show that Acrylic-IOL are not appropriate for a Dynasilan® -coating.
