Effect of Prostaglandin F2a on Proliferation and Collagen I Expression in human Keratocytes in vitro

Purpose: Latanoprost,a phenyl- substituted synthetic analogue of prostaglandin (PG) F2a- isopropylester, has been shown to be an effective intraocular pressure lowering agent in normal, ocular hypertensive and glaucomatous eyes. The pressure lowering effect of Latanoprost eye drops may be caused by a reduction of collagen I expression within the uveoscleral outflow pathway. The modulation of cell proliferation and collagen I synthesis in various tissues are major effects of PGF2a. However, up to now it remains unknown whether PGF2a modulates also the proliferation and collagen I expression in human keratocytes.
Methods: Human keratocytes (HK) were cultured according to established protocols. Experiments were performed with cells of the 1. and 2. passage under serum reduced conditions. 104 cells /well were seeded and cells were then incubated with different (5 x10-5 - 5 x10-3 mg/ml) concentrations of PGF2a (Sigma Ind.).The number of cells was determined after 24, 72 and 120 hours. The modulation of collagen I expression was tested in PGF2a treated cultures by immunofluorecences cytochemistry using fetal mouse anti - collagen I antibodies.
Results: Depending on the concentration PGF2a induced a significant stimulation of HK proliferation(more then a 2 fold increase in cell number at the highest concentration of PGF2a after 120 hours).The collagen I expression was reduced in PGF2a treated cell cultures in comparison to unsupplemented control cultures.
Conclusion: For the first time the present study demonstrates the effect of PGF2a on proliferation and collagen I expression in HK. It appears interesting that the number of cells increases but the expression of collagen decreases in PGF2a treated cell cultures.The reduction of collagen could be an answer to the thinning of the cornea after prolonged use of Latanoprost eye drops described in other studies before.
