Analysis of chromosomal breaks in cultured human keratocytes after excimer-laser photoablation

Purpose: To evaluate the rate of chromosomal breaks in cultured human keratocytes after excimer-laser photoablation.
Material and methods: Three pairs of human corneas which were not suitable for therapeutic uses were investigated. One of each pair underwent excimer-laser photoablation including the epithelium, bowman´s layer and anterior stroma (193 nm, 180 mJ/cm², 4Hz). The second cornea of each pair served as control. After isolation, the keratocytes were cultured using Amniomax®. Keratocytes were stained with giemsa and GTG and were analysed for chromosomal breaks performing ISCN (5 analysis per culture).
Results: The rate of chromosomal breaks in keratocytes of corneas which underwent photoablation was 0.16/cell (SD=0.06) compared to 0.12/cell (0.04) in keratocytes from corneas which served as controls (p>0.05).
Conclusion: There was no statistically significant difference in the rate of chromosomal breaks in keratocytes from human corneas which underwent excimer-laser ablation compared to keratocytes from corneas which served as controls.
