Amiodarone: functional and morphological examination in the retina of mice

Objective: The purpose of this study was to evidence and localize lipidosis-like alterations caused by amiodarone, a cationic-amphiphilic drug, in the retina of mice by means of electroretinography compared with histological investigations.
Methods: Pigmented mice of the stock Black CL 57 were feeded with amiodarone (average 176 mg/kg body weight) for 22 weeks. The ERG measurements were done initially and after 22 weeks. Afterwards the retinae were prepared for the histology.
Results: After 22 weeks there were no significant changes in the following electroretinographic parameters: a-wave amplitude, b-wave amplitude, awave time to peak and b-wave time to peak. The histological preparations of the retinae showed no lipidosis-related inclusions in the light microscope.
Conclusion: The standard-ERG used in this study seems not to be suitable to detect functional changes in the retina caused by amiodarone. To prove minimal lysosomal inclusions for example in the pigment epithelium or in the ganglion cells you need more sensitive and more specific leads.
