Expression of PDGF-receptor type beta in human tenon's capsule fibroblasts after stimulation with cytokines in vitro

Previously we showed, that PDGF isoforms are the most potent stimulators of proliferation of human tenon's capsule fibroblasts (HTF) in vitro. The aim of the present study was to investigate, whether treatment of HTF with different cytokines modifies the expression of the corresponding cell surface receptor PDGF receptor type beta (PDGFR-ß). Second passage HTF were stimulated for six days with the following cytokines at respective concentrations (PDGF-AA 0,01 - 100 ng/ml, PDGF-AB 0,01 - 100 ng/ml, PDGF-BB 0,01 - 100 ng/ml, EGF 0,01 - 100 ng/ml and IL-1 ß 0,001 - 10 ng/ml). The expression of PDGFR-ß was detected using polyclonal antibodies in the Western Blot and analysed semiquantitatively with densitometric techniques. The analysis of the immunoblot results demonstrated a significant reduction of expression of PDGFR-ß after stimulation with 100 ng/ml PDGF-AB and 100 ng/ml PDGF-BB. In contrast, the remaining cytokines did not alter expression of PDGFR-ß even at high concentration levels. Our results suggest, that high concentrations of PDGF-AB and PDGF-BB lead to a reduced expression of PDGFR-ß during wound healing, thus modifying cell proliferation in the wound region.
