Immunohistopathologic Evaluation of Proliferative Activity in Choroidal Neovascularization after PDT

Purpose: To evaluate the proliferative activity of classic choroidal neovascular membranes (CNVM) secondary to age related macular degeneration (ARMD) excised at different times (3 days- 5 months) after photodynamic therapy (PDT). Method: Seven subfoveal classic CNVM, excised between 3 days and 5 months after PDT, were stained for CD34 (panendothelial marker), CD105 (proliferation marker for endothelial cells), and Ki-67 (nuclear marker of proliferation) and compared to the clinical course. Results: Fluorescein angiography (FA) performed on the day of surgery disclosed non-perfusion of the treated area 3 days after PDT, but reperfusion and leakage was seen at greater post-PDT intervals. Immunohistopathologic examination of 4 CNVM excised 3 days after PDT disclosed several vessels that were positive for CD34 and CD105. The endothelial cells appeared damaged, but several of the lumina were not occluded. Ki-67 expression was prominent only in one specimen.
