Paraffin Embedding Technique for Specimens Obtained by Vitrectomy

Purpose: Histological and immunohistochemical evaluation of vitrectomized specimens is challanging, since paraffin embedding and long time storage of such a material is not possible. In this study we introduce a technique for paraffin embedding of vitrectomized specimens.
Method: Vitrectomized material was collected in 125 cases. The material was centrifuged at 2000 rpm, formed to a hard block using thromboplastin, dehydrated and then routinely embedded in paraffin.
Results: Paraffin embedding of vitrectomized material was easy and fast to perform. Routine histologic examination of the tissue including 4µm thick sections, immunohistochemical staining and tissue storage for later preparation could be done.
Conclusions: Paraffin embedding of vitrectomized material is possible using the block forming technique with thromboplastin. This technique gives us for the first time the possibility to embed vitrectomized material and cell cultures as a paraffin block including the advantages of immunohistochemical staining for different antibodies and long time storage.
