Purpose: To study the influence of gender and age on iris colour characteristics of healthy adults in a cross-sectional study with a newly developed method for automatic digital image processing. Method: In a cross- sectional study 4000 iris photographs were obtained from 2000 adults aged 40 to 80 years under standardized conditions by analogue photography and then digitised by standardized colour scanning. Customized automatic pattern-recognition software was used to identify iris structures and to obtain colorimetric and geometric iris data. Data was analysed separately for upper, lower, left and right quadrant of each iris. Digital colour-information was used for statistical analysis of RGB mean vector and other parameters. To assess the influence of age and sex on mean iris colour and differences between nasal and temporal iris-quadrant a full factorial repeated measures analysis of variance with between-factors gender, age (decades) and within-factors quadrant (nasal, temporal), colour channel (RGB) was performed. Results: We included 1096 women (58.5±10.4 years) and 868 men (61.3±10.2 years) in the primary statistical analysis (one random eye per participant). All factor main effects (between and within
