Inhibition of PDGF-BB-induced Proliferation of Cultured Human Tenon's Capsule Fibroblasts by the PDGF-receptor Specific Tyrphostin AG-1295

Purpose: PDGF-BB is currently thought to be one of the most potent stimulators of cell proliferation during wound healing after glaucoma filtering surgery. In the present study we investigated the antiproliferative effects of the tyrphostin AG-1295, a PDGF-receptor-ß-specific tyrosin kinase inhibitor, on cultured human tenon's capsule fibroblasts (HTF) after stimulation with PDGF-BB and 10% FCS. Method: Cultured HTF were treated with increasing concentrations of AG 1295 (0.1, 1, 10 and 100 mmol/l) and DMSO (0.001%, 0.01%, 0.1% and 1%) as a control while cell proliferation was stimulated with PDGF-BB (50ng/ml) and fetal calf serum (FCS, 10%). The cells were harvested with trypsin/EDTA on days 1, 3, 5, 8, 10 for counting. Results: PDGF-BB- and FCS-induced cell proliferation of HTF was found to be significantly reduced, when HTF were simultaneously treated with high concentrations of AG-1295 (100 mmol/l). Inhibition by AG-1295 (100mmol/l) was significant after 3 days (77 %), 5 days (157%), 8 days (126%) and 10 days (121%), when HTF were stimulated with PDGF-BB. Furthermore, inhibition by AG-1295 (100mmol/l) was significant after 8 days (65%), when HTF were stimulated with FCS 10%. Conclusions: The result
