In vitro Cornea Model Corneal Re-epithelisation after Mechanical Trauma

Purpose: The avascular cornea is exposed to external influences and is vulnerable to chemical irritation. The biological response to such traumas provoces changes in cellular function within the cornea and the release of inflammatory mediators that initiate a secondary inflammatory response in the adjacent conjunctiva. According to animal experiments we attempt to demonstrate that the ex vivo cornea offers a suitable model to investigate external eye damage.
Method: We designed a PLEXIGLAS® perfusion chamber to clamp corneas to a horizontal convex device, suitable for topical agent delivery, offering suitable conditions for investigate isolated rabbit cornea. Regarding the corneal regeneration we placed a standardized 6 mm corneal epithelial defect by hockey knife abrasion on 4 corneas. Epithelial healing was documented by fluoresceine stains and photographic documentation.
Results: Complete epithelial regeneration was observed between 7 and 9 days.
Conclusions: We establish a perfusion chamber to form a reliable tool for in vitro examinations of epithelial regeneration and toxicity tests in ex vivo isolated corneoscleral
