Ocular Expression of Noelin, a Member of the Olfactomedin Family

Purpose: Mutations in myocilin are causative for some forms of primary open angle glaucoma. Most of these mutations are localized in the olfactomedin domain of myocilin, which is highly conserved during evolution. Noelin is a member of the same protein family, which has an olfactomedin domain with high homology to that of myocilin. Since noelin might share important functions with myocilin, we investigated its expression in human and rat eyes. Method: MRNA expression of noelin in rat ocular tissues at various ages as well as in human ocular tissues was investigated by northern blot analyses and RT-PCR. PCR products were cloned, sequenced and used as probe for northern blot analyses. Trabecular meshwork (TM) cells in culture were incubated with TGF-b1 [1nM/ml] and dexamethasone [10-7M] for 3 days. Results: In rat tissues, the strongest hybridization signal for noelin was detected in the retina. A significant lower expression was observed in the sclera and the optic nerve. By RT-PCR of human ocular tissues, a specific band for noelin was seen in the retina, sclera, cornea and TM.
