SupraDescemetic Synthetic Cornea (sDSC) Preliminary Results of a Novel Non-penetrating Keratoprosthesis in the Rabbit Model

Purpose: To show feasibility and biocompatibility of a novel non-penetrating keratoprosthesis (supraDescemetic Synthetic Cornea, sDSC) in a rabbit model. Method: A new surgical technique was developed to implant the keratoprothesis (7 mm diameter, 450mm thick optical zone) on top of a bare Descemets membrane (DM). 4 different materials were tested in this series: hydrophobic PMMA, hydrophilic pHEMA-MMA26, pHEMA-MMA34 and pHEMA-NVP75. The device was successfully implanted in healthy eyes of 17 New Zealand White rabbits and followed for up to 7 months to date. 7 rabbits were selected for histological evaluation. In 16 rabbits an ocular surface injury was created by application of 1-heptanol onto the cornea and surgical removal of the limbus, leading to vascularisation and scarring.
